
Reconstitution Best Practices for Lyophilised Research Peptides: mL, mg/mL, U-100 Syringe Conversions
TL;DR. Research-grade lyophilised peptides are typically reconstituted in bacteriostatic water (0.9% benzyl alcohol) at 1-2 mL per vial. The working-stock concentration (mg/mL) determines both the dose volume and the U-100 syringe unit count for administration. This article covers the standard conventions plus stability windows.
The reconstitution equation
Working-stock concentration = vial mass (mg) ÷ reconstitution volume (mL)
Example: A 10 mg vial + 2 mL bacteriostatic water = 5 mg/mL working stock.
Dose volume = desired dose (mg) ÷ working stock (mg/mL)
Example: 0.5 mg dose ÷ 5 mg/mL = 0.10 mL per administration.
U-100 syringe unit count = dose volume (mL) × 100
Example: 0.10 mL = 10 units on a U-100 syringe.

Reconstitution medium choice
- Bacteriostatic water (0.9% benzyl alcohol), 28-day stability at 2-8 °C. Standard choice.
- Sterile water, 24-hour use only; no preservative.
- PBS pH 7.2, for cell-culture applications; verify peptide is stable at neutral pH.
- DMSO / DMF, for hydrophobic peptides that don't dissolve in water. Verify with the CoA.
Never use reducing-agent-containing buffers (DTT, BME) for peptides with disulfide bonds, they'll cleave the disulfide.

Common reconstitution mistakes
- Injecting water directly into the powder cake, creates foaming. Correct: inject slowly against the vial wall.
- Vortexing to dissolve, can denature the peptide. Correct: gentle swirling, allow 5-10 min at room temp.
- Using expired bacteriostatic water, check the water expiration date; benzyl-alcohol preservative degrades.
- Wrong syringe reading, a U-100 insulin syringe has 100 units per mL; a U-40 has 40 units per mL. Always verify the syringe label.
